Suite à une gaffe monumentale, deux jeunes policiers sont assignés à l'unité « 21 Jump Street », qui est chargée d'infiltrer une école secondaire où sévit un réseau de trafiquants d'une nouvelle drogue particulièrement dangereuse. Pour Schmidt et Jenko, ce sera l'occasion de recommencer différemment une étape importante de leur vie d'adolescents et de réparer certaines erreurs. Mais leur nonchalance risque de compromettre leur mission, d'autant que Schmidt s'est lié d'amitié avec une étudiante qui s'avère être la petite amie du dealer de l'école.
